Abstract:
The invention provides a nucleic acid molecule comprising an mRNA transcript comprising a reporter gene coding sequence operably linked to a fragment of a human vascular endothelial growth factor (VEGF) 5' untranslated region (UTR) encoded by the sequence of SEQ ID NO:3 (sequence 330 -1080 of the 5'UTR, and a human VEGF 3' UTR, wherein the fragment of the human VEGF 5' UTR is upstream of the reporter gene coding sequence and the human VEGF 3' UTR is downstream of the reporter gene coding sequence, and wherein the mRNA transcript does not comprise a sequence encoded by SEQ ID NO:9 (sequence -7 - 470 of the 5'-UTR) or a fragment thereof. The invention further provides a vector comprising the nucleic acid molecule of the invention, a method of expressing the nucleic acid molecule as well as the use of the nucleic acid molecule for screening a group of candidate compounds for a compound that modulates UTR-dependent translation of human VEGF mRNA.
