Abstract:
To provide equipment that can be assembled at about one-fourth of the cost by improving separation ability using flash chromatography of a linear gradient type of flash chromatography instead of an expensive device such as high-performance liquid chromatography for separation, by which necessary parts can be easily procured.SOLUTION: Whereas a gradient method for linearly changing the eluent concentration uses two solutions as the suction from the lower column and the feed pressure from the upper column, flash chromatography of good linear gradient method was completed by making a first bottle in a pressure-sucking state and a second bottle in a micro-hole open state. Also, when performing fractionation operation in the apparatus, generation of bubbles from the lower column became the cause of measurement errors. The ratio of the suction from the lower column and the feed pressure from the upper column was precisely investigated to solve this problem, and the suction force 0.8 and the liquid feed pressure 1.0 were found.SELECTED DRAWING: Figure 2
