To cleave the Cα-C bond and side chain of the peptide backbone to detect the product with high sensitivity.SOLUTION: The cleavage method is a cleavage method that specifically cleaves the Cα-C bond and/or side chain of the peptide main chain, and includes: preparing a sample containing a molecule having a peptide main chain in which multiple amino acids are peptide-bonded to each other; and performing laser irradiation of the sample in the presence of at least one of 3,5-dihydroxy-2-nitrobenzoic acid and 3,5-dihydroxy-4-nitrobenzoic acid.SELECTED DRAWING: Figure 5
