Abstract:
To provide a method for purifying or regenerating a chromatography material for reuse in large scale production of a plurality of polypeptide products.SOLUTION: A method includes the steps of: a) allowing a chromatography material to pass through 2 material volume or more of elution buffer, in which the elution buffer contains approximately 0.15 M of acetic acid and has approximately pH 2.9; b) statically holding the material in the elution buffer over a period of time within a range from approximately 10 minutes to approximately 30 minutes; c) allowing the material to pass through approximately 2 material volume or more of elution buffer; and d) allowing the material to pass through approximately 2 material volume or more of regeneration buffer, in which the regeneration buffer contains approximately 0.1 N of NaOH and has approximately pH 13.SELECTED DRAWING: Figure 7
