Abstract:
PURPOSE:To provide a human carcinoma necrosis factor polypeptide essentially free from impurities, and separated and purified from a product containing human carcinoma necrosis factor and produced by a genetic recombination technique. CONSTITUTION:A DNA coding the human carcinoma necrosis factor polypeptide and its precursor is separated. A start codon ATG is added to the 5'-terminal of the DNA base sequence coding said polypeptide, and a DNA fraction having a base sequence containing the termination codon at the 3'-terminal is prepared, and integrated into a vector to obtain a non-fused plasma manifestation vector. The vector is introduced into a proper host, and the obtained transformation is cultured. The crude product separated from the cultured product is purified by conventional protein purification process to obtain a polypeptide having L-M cytotoxicity in vitro and antitumor activity to Meth-A sarcoma in vivo. The protein has the amino acid sequence of formula, a molecular weight of 45,000+ or -5,000 dalton in the absence of urea and 18,000+ or -3,000 dalton in the presence of urea by gel-filtration method, and an isoelectric point of 5.9+ or -0.3.
