Abstract:
The invention relates to a method to optically focus on a target layer in a microplate well defined in a microplate, said well having a transparent region below the well, the method comprising: a) optically sensing, using specular reflection from a beam having a focal spot, an optical reference point at a specified location on an underside of a microplate; b) moving the focal spot of the beam a fixed distance from said reference point to a target layer in the well at least partially above the bottom of the microplate well; and c) repeating steps a)- b) for a plurality of locations, corresponding to different microplate wells.
