To provide a method for amplifying a nucleic acid by SmartAmp2 by which the amplification rate of the nucleic acid can be controlled.
The method for amplifying the nucleic acid uses the following first primer and second primer. The first primer contains, at the 3' terminal, a sequence (Ac') hybridizing with 3' terminal of a sequence (A) of a target nucleic acid sequence, and contains, at the 5' side of the sequence (Ac'), a sequence (B') hybridizing with 5' side of a complementary sequence (Bc) of a sequence (B) of the sequence (A), in the target nucleic acid sequence. The second primer contains, at the 3' terminal, a sequence (Cc') hybridizing with 3' terminal of the sequence (C) at the complementary sequence of the target nucleic acid sequence, and contains, at the 5' side of the sequence (Cc'), a folded sequence hybridizing with the same strand, and controls at least one of a melting temperature and a free energy of the folded sequence.
