Abstract:

To provide a method for producing a plant body containing high-concentration tryptophan by a gene-targeting method, and the plant body and the like produced by the method.

In designing a vector used for a gene-targeting method, a point mutation of S126F is brought to locate in the right border (RB) side. By arranging the S126F mutation in the RB side of the T-DNA, it is confirmed that a gene-targeting individual is efficiently obtained even in the case when a homologous sequence region must be short. It is found that it is effective to use 5MT in 500 μM which is higher than the concentration ever reported in selecting an individual succeeded in gene-targeting of OASA2 gene. The gene-targeting individual thus obtained accumulates about 8.62 nmol/mg isolated tryptophan in rice leaf blades. It is about 130 times the content of wild type isolated tryptophan.
