Abstract:
PROBLEM TO BE SOLVED: To provide an image acquisition apparatus capable of securing the amount of light during imaging and accurately detecting a focal position of a sample, and a focusing method for the same.SOLUTION: An image acquisition apparatus M can form an optical path length difference of a second optical image without branching of light in a second optical path L2, by the use of an optical path difference generating member 21. Thus, the amount of light to the optical path L2, needed to acquire information on a focal position, is suppressed to secure the amount of light during imaging with a first imaging device 18. The image acquisition apparatus M synchronizes movement of a predetermined portion of a sample S in the field V of view of an objective lens 15 and rolling-readout so that an optical image in the predetermined portion of the sample S is exposed at each pixel array 20b of a second imaging device 20. The acquired image data includes contrast information equivalent to that when the focal position of the objective lens 15 is changed in the same portion of the sample S, and based on the information, focal point information can be calculated quickly and accurately.
