Abstract:
1291644 Substituted 1-indanecarboxylic acids, esters and salts BRISTOL-MYERS CO 30 Jan 1970 [31 Jan 1969] 4535/70 Heading C2C The invention comprises d-, l- and dl-compounds of formula and nontoxic pharmaceutically acceptable salts and C 1-6 alkyl and (up to C 8 ) tertiary amino alkyl esters thereof, wherein Y is H, Cl, Br, F, OH, C 1-6 alkyl, alkoxy or alkylthio, SH, CN, NO 2 or NH 2 . These compounds are prepared by reacting p - cyclohexylphenylsuccinic anhydride [from the acid, itself made via p-cyclohexylbenzaldehyde, diethyl p-cyclohexylbenzylidenemalon. ate and ethyl 3-cyano-3-(p-cyclohexylphenyl)- propionate] with a Friedel-Crafts catalyst in an inert organic solvent to give 5-p-cyclohexyl-3- oxoindan-1-carboxylic acid, and reducing the latter, and where desired producing the compounds where Y is other than H by standard substitution methods (optionally preceded or followed by esterification using the acid or acid chloride); including nitration, reduction of the nitro-compound and utilization. of the intermediate where Y is a diazonium group (II). Circhonidine and dehydroabietylamine salts are used in examples for optical resolution. The compound where Y=HS may be prepared by saponifying the intermediate in which Y= EtOCS 2 -(obtained from II). The intermediate of the above formula where Y=I is also mentioned. Therapeutic compositions for oral or parenteral administration comprise compounds of the above formula, which are anti-inflammatory agents.
