Abstract:

PROBLEM TO BE SOLVED: To obtain a new 1-azidoindan-2-ol derivative useful as an intermediate for producing variety of physiologically active substances, etc.
SOLUTION: This new 1-azidoindan-2-ol derivative comprises both trans-1- azidoindan-2-ol derivative of formula I, e.g. (±)-trans-1-azidoindan-2-ol, and cis-1-azidoindan-2-ol derivative of formula II, e.g. (+)-(1S, 2R)-cis-1-azidoindan-2- ol. The compound of formula I is, for example, obtained by reacting indene with N-bromosuccinimide and forming a bromohydrin in an aqueous terahydrofuran, further forming an epoxide by a ring-closing reaction of the bromohydrin by using an alkali such as NaOH and reacting the resultant epoxide with an azide compound. Further, an optical resolution of the compound of formula I is performed by a transesterification reaction thereof with a fatty acid vinyl ester by using a lipase to provide the compound of formula II. The reaction is performed by using an organic solvent not inhibiting the activities of the lipase at 10-100°C for 1-300hr and a hydration, etc., of the resultant product is performed under a basic condition.
