Abstract:
PURPOSE: To provide new 1-cyanomethyl-4-carboxymethyl-2-ketopiperadine and its salt useful as an intermediate for synthesis of ethylenediaminetriacetic acid or its salt.
CONSTITUTION: This compd. is represented by the formula (where R is H, alkali metal or alkaline earth metal), e.g. 1-cyanomethyl-4-carboxymethyl-2- ketopiperazine. The compd. of the formula is obtained by reacting formaldehyde with ethylenediaminediacetic acid or a salt contg. a base cation of ≤1 equiv. in aq. soln., and then reacting with a cyanide source. In the first step of a two step reaction, the reaction is preferably proceeded at about 15-65°C and in the second step it is at a room temp. to about 70°C. As a cyanide source, hydrogen cyanide, metal cyanides are preferable, and it is preferable to acidify ethylenediaminediacetic acid with an acid of ≤3 in pKa.
