Abstract:

To obtain a new compound which is a specific 2,8- dioxabicyclo[3.2.1]octane derivative having a squalene synthetase inhibiting action, effective for suppressing the biosynthesis of cholesterol without causing side action and useful as an agent for the treatment of hyperlipemia, etc.

The objective new 2,8-dioxabicyclo[3.2.1]octane derivative is expressed by formula I [R1 is a (substituted)2-10C alkyl or alkenyl; R2 is H, a (substituted)2-10C alkanoyl or alkenoyl; R3 is H, a 1-5C alkyl or a (substituted) arylmethyll, has squalene synthetase inhibiting action and is useful e.g. as an agent for the treatment of hyperlipemia. The compound can be produced from a glucofuranose derivative of formula II [R4' to R7' are each a (substituted)alkyl, an arylmethyl, silyl or an acyl] by a multi-step synthetic reaction consisting of a combination of reduction, selective oxidation of hydroxyl group, hydroxyalkylation, cyclization, etc.
