PURPOSE: To obtain the unaggregated tissue plasminogen-activating factor (tPA) in a high yield by discharging a tPA-containing effluent from a cell culture tank containing the cells expressing the tPA, while the unaggregated tPA is contained in a larger amount than the aggregated tPA.
CONSTITUTION: A tPA-containing effluent is discharged from the cell culture tank containing cells expressing tPA, while the unaggregated tPA is contained in a substantially larger amount than the associated tPA. The discharged tPA- containing effluent is treated under a condition substantially capable of preventing the aggregation of the unaggregated tPA, thereby producing the unaggregated tPA from the culture cells expressing the tPA in a high yield. The cells expressing the tPA preferably comprises mammalian cells expressing the tPA encoded in a recombined DNA-expressing vector. The discharged tPA- containing effluent is preferably treated at a temperature of 0-15°C.
