Abstract:
PURPOSE: To obtain a novel steroid useful as a versatile intermediate for active type vitamin Ds which improve bone mass.
CONSTITUTION: A steroid of formula I [R1 is CH2OH, (CH2)3OH or the like; R2 is a protecting group; R3 is an alkoxy or the like], for example, 1α,3β- dihydroxy-2β-(3-hydroxypropyl)-23,24-dinor-5,7-choladiene-22-ol 1α, 2β(3-),3β- tristetrahydropyranyl ether. The compound of formula I is obtained by epoxidation of the compound of formula II [R'1 is CH2OR4 (R4 is a protecting group) to derivate into 1α,2α-epoxide, elimination of R2 from the product to change into an epoxy alcohol, reaction of the epoxy alcohol with alcohol to change into a compound of formula III (R2 is R3), protection of the OH in the product to obtain compound of formula IV, elimination of the protecting groups for 5,7-diene and R4, oxidation of R1, when needed followed by conversion of the hydroxymethyl on R1 into an aldehyde.
