Abstract:

PURPOSE: To enhance enzymatic biosyntheses at a low cost by bringing a cytoclasis preparation of an organism capable of producing ≥1 species of penicillin, cephalosporin and cephamycin to act on an amino acid mixture.

CONSTITUTION: Cephalosporium Aceremonium cells are extracted and stabilized with glycerol to obtain a cell-free extract (A). The component A is added to a mixture (B) of L-α-aminoadipic acid, L-cysteine and L-valine (ACV) and an ACV synhetase is reacted. Each amino acid in the ACV is successively bonded to transform to isopenicillin of formula I, then penicillin N of formula II and then desacetoxycephalosporin C of formula III [X is H(4a)]. If needed, an ACV synthetase derived from eucaryotic β-lactam-producing organisms is reacted to the ingredient A to produce a β-lactam antibiotic.
