PURPOSE: To produce aequorin fused with a protein having antibody-bonding ability by fusing a protein having specific bonding ability (or its gene) with aequorin (or its gene).
CONSTITUTION: The figure shows the construction steps of manifestation vector of aequorin fused to protein A. The process is carried out as follow color. A fragment of aequorin cDNA is separated from aequorin-manifestation vector piPHE by digesting with EcoRI. The fragment is inserted into the EcoRI site of a protein A fusion manifestation vector pRIT5 to obtain pAAQ1. The pAAQ1 is digested with SmaI/PvuII and subjected to self-ligation to obtain pAAQ2. It can be applied to the detection with luminescence by utilizing the specific bonding ability and luminescent nature of the fused protein. The fused protein can be produced in a mass by the use of E.coli, etc., as the host.
