Abstract:
PURPOSE: To obtain an amide derivative, having functions to inhibit activities against calpain and useful for treating intractable diseases.
CONSTITUTION: This compound of the formula R-A-CO-NH-CH(CHO)-(CH2)n-B [R is a substituted amino of formula I (X is CH, or N; Y is CH2, CO, SO2, NHCO, etc.; W and Z are each H or together CO or SO2); A is a cycloalkylene; n is 0-5; B is a lower alkyl, an aromatic hydrocarbon or a heterocyclic ring], e.g. cis-4-((2-naphthoyl)amino)-1-cyclohexanecarboxylic acid (1R,S)-(1-formyl-2- phenyl)ethylamide. The compound is obtained by condensing a carboxylic acid derivative of the formula R-A-COOH with an amino-alcohol derivative of formula II, providing an alcohol derivative of the formula R-A-CONH-CH (CH2OH)-(CH2)n-B and subsequently oxidizing the resultant alcohol derivative.
