PURPOSE: To attain a high speed and high precision in analysis by a method wherein light information on a cell is subjected to coordinate transformation into two parameters and an area of a part obtained by division by one parameter is divided further by using a histogram of the other parameter.
CONSTITUTION: When red fluorescence and green fluorescence of detected blood are taken for the axis (x) of abscissa and the axis (y) of ordinate respectively, for instance, cell groups corresponding to a neutrophil, a lymphocyte, a monocyte, an eosinophil and a basophil respectively appear on a scatter diagram. Then, coordinate transformation is conducted so that each cell group is contained in an area obtained by division only by straight lines being horizontal in relation to the axis (x) and the axis (y). Next, a minimal point on a histo-paragram is determined first by using a threshold value y1 as a fixed value, threshold values x1, x2, y1 and y3 for dividing the cell groups from each other are determined sequentially, and by using these values, the area is divided into areas L, B, M, E and N containing the lymphocyte, the basophil, the monocyte, the eosinophil and the neutrophil respectively. Then cell data in the areas are counted and compared and thereby the ratio of the kinds of white blood corpuscles with one another is determined. According to this method, a high-speed and accurate analysis can be executed automatically.
