PURPOSE: To obtain the subject antibody, produced by a hybridoma prepared by using a human-derived tissue factor as an immunogen and useful as an immunosuppressant, etc., for various diseases, etc., due to cellular hyperimmunization in which rejection to organ transplantation or an activated macrophage participates.
CONSTITUTION: The objective monoclonal antibody, specific for a human-derived tissue factor, belonging to the IgG2a class and IgM class and capable of recognizing a specific marker tissue factor of an activated macrophage is obtained by administering a human- derived tissue factor as an immunogen together with an adjuvant to a BALB/c mouse, immunizing the mouse, collecting a cell of the spleen after the final immunization, fusing the collected cell of the spleen to a cell of a murine myeloma, then culturing the fused cell in an HAT culture medium, providing a hybridoma, screening the supernatant of the culture solution of the hybridoma by an enzyme-linked immunosorbent assay(ELISA) method, selecting a strain capable of producing an antibody, cloning the resultant strain according to a limiting dilution method and culturing the monocloned cell. The IgG2a antibody has the cross-reactivity toward a human placenta- derived tissue factor and the IgM antibody has the cross-reactivity toward a canine and a rabbit brain-derived tissue factors.
