Abstract:
PROBLEM TO BE SOLVED: To provide a method for producing antibody-agent conjugate having high agent loads, and to provide a method of use.SOLUTION: A transglutaminase-mediated antibody-agent conjugate having high agent-antibody ratio (DAR) contains 1) endogenous glutamine which is made reactive by engineering glutamine-containing tag, endogenous glutamine, and/or antibody or engineered transglutaminase (e.g., by change of substrate specificity); and 2) an amine donor substance containing an amine donor unit, a linker, and an active substance part; wherein DAR is at least about 5.
