Abstract:
PROBLEM TO BE SOLVED: To provide antisense compositions and methods of making and using same.SOLUTION: The present invention provides pharmaceutical formulations for oral administration of antisense oligonucleotides, such as antisense oligonucleotides against SMAD7. The pharmaceutical formulations can be used to treat Crohn's disease, ulcerative colitis and chronic inflammatory bowel disease. In one embodiment, a pharmaceutical formulation for oral administration of an antisense oligonucleotide that comprises an intra-granular phase is provided, wherein the intra-granular phase includes an antisense oligonucleotide such as that represented by SEQ ID NO: 1, or a pharmaceutically acceptable salt thereof (such as a sodium salt), and a pharmaceutically acceptable filler, and the pharmaceutical formulation may also include an extra-granular phase, that may include a pharmaceutically acceptable excipient such as a disintegrant.
