Abstract:

PROBLEM TO BE SOLVED: To enable a highly clear and accurate assay by a small amt. of a reagent by directly adding a particle label simultaneously sensitized to non- specific protein capable of being participated in the control reaction of a first specific binder and a separate specific binder.
SOLUTION: A particle label (e.g; latex particles having a proper size) is brought into contact with a mixture of two materials for sensitizing the particles in an aq. suspension to prepare a simultaneously sensitized particle label. For example, it is necessary to simultaneously sensitize a mixture of a mouse monoclonal antibody to the α-chain of hCG in a proper specific wt. ratio and a non-specific polyclonal antibody (e.g; rabbit IgG) under a buffering condition in the same way as the case of standard procedure. After a time interval sufficient to bond these materials on latex particles, the unbonded material is separated by a conventional method such as centrifugal separation and the simultaneously sensitized particles can be re-suspended in a form of a storage buffer soln. usuable in a test as it is.
