PURPOSE: To perform efficient affinity separation by chemically bonding epoxy groups to the surface of a membrane of polyolefin or an olefin-olefin halide copolymer and the surfaces of the pores in the membrane.
CONSTITUTION: A hydrophobic membrane of polyolefin or an olefin-olefin halide copolymer is irradiated with ionized radiation, preferably electron beams or γ-rays in vacuum or in an inert gas. Vinyl acetate is grafted on the irradiated membrane in a vapor phase to bond polyvinyl acetate to the surface of the membrane and the surfaces of the pores in the membrane. The grafted polyvinyl acetate is modified to polyvinyl alcohol by saponification. Haloepoxide, preferably epichlorohydrin or bifunctional epoxide is then introduced into the membrane in an aprotic polar org. solvent to obtain a base membrane for an affinity separation membrane fit to adsorb, separate or purify protein, an amino acid, an antigen, an antibody, enzyme, etc., selectively and more efficiently than chromatography.
