PROBLEM TO BE SOLVED: To obtain a new compound reducing a high cardiac filling pressure occurring in heart failure to thereby lessen a burden to heart because of a neutral endopeptidase inhibitory activity, enhancing a uretic function and useful for the heart failure.
SOLUTION: This invention relates to a compound of formula I (R1 is e.g. a phenyl-lower-alkyl or a naphthyl-lower-alkyl; A is e.g. CH2 or O; R2 and R3 are each H or a halogen; and R4 and R5 are each H or a bioactive ester- forming group) and its salt, e.g. (3S, 2'R)-3-{1-[2'-(ethoxycarbonyl)-4'-phenyl- butyl]-cyclopentane-1-carbonylamino}-2,3,4,5-tetrahydro-2-oxo-1H-1-benzazepine-1- acetic acid. The compound is obtained by reacting an acid of formula II (R4a is an acid protecting group) or its derivative with an amine of formula III (R5a is an acid protecting group) and, if necessary, deprotecting the resultant amide.
