Abstract:

PURPOSE: To supply a mammalian T-cell receptor in a large amount, by cultivating a microorganism containing a recombinant DNA, having a DNA capable of coding a β-chain constant-region polypeptide of a specific mammalian T-cell receptor and integrated thereinto.

CONSTITUTION: A microorganism containing a recombinant DNA having a DNA capable of coding a β-chain constant region polypeptide of a mammalian T-cell receptor expressed by formula I is cultivated. A polypeptide is accumulated in the resultant culture by this cultivation and then the aimed β-chain constant-region polypeptide of the mammalian T-cell receptor is collected. In this case, a plasmid having a tryptophan promoter is used and the DNA is preferably integrated into the downstream side of the tryptophan promoter. Furthermore, a microorganism belonging to Escherichia coli is preferred as the microorganism.
