Abstract:
NEW MATERIAL: A compd. of formula I [R1 is H, OH; methyl and HO at 6- and 7-positions respectively can be either in α or β configuration).
EXAMPLE: 3-α-7-Dihydroxy-6-methyl-5-β-cholan-24-oic acid.
USE: The compd. is useful for the treatment of cholelithiasis as an antimaldigestion agent, digestion promotor, antidyslipidosis and bile secretion promotor.
PROCESS: A compd. expressed by formula II (R2 is an OH protecting group; R1' is H, an OH protecting group) is methylated under a kinetic control condition and in succession the resultant compd. is reduced by deprotection. The methylation is preferably executed by using methyl iodide in the presence of lithium and a dialkylamide in a solvent, such as 1,2-dimethoxyethane, THF, ethanol.
