PURPOSE: To prepare bovine embryonic stem cells which can be subcultured in a large volume and can be used as a material for transduction of exotic gene or bovine ontogenesis.
CONSTITUTION: The inner cell mass and trophectoderm are taken out of a blastocyst originating from bovine fertilized ovum and cultured on the inactivated feeder cells in a medium containing at least fetal calf serum, 2- mercaptoethanol, leukemia control factor and basic fibroblast growth factor, and the embryonic stem cells are isolated and collected from the cell culture mixture. The embryonic cells show morphologically the same shape as the inner cell mass of the blastocyst and the mitotic image shows XY or XX type diploid cell nucleus under microscopic observation and the X-chromosomes are activated to express SSEA-1 or LEx antigen and their subculture is possible.
