Abstract:
PROBLEM TO BE SOLVED: To provide a gene expression cassette for stable and high production of a target protein.SOLUTION: The present invention provides a gene expression cassette having a structure in which a DNA construct (X) containing a gene to be expressed and a poly-A addition sequence is sandwiched between a promoter (P) and an enhancer (P'). The gene expression cassette further comprises a transposon sequence (T) upstream of the promoter (P) and downstream of the enhancer (P'). Furthermore, in the gene expression cassette, by appropriately arranging a nuclear matrix-binding sequence (M) upstream of a replication-initiating sequence (S) in combination with the transposon sequence (T), it is possible to more effectively produce the target protein in large quantity in a stable manner. For example, histidine-rich glycoprotein (HRG) and antibodies can be produced in large quantities in a stable manner.SELECTED DRAWING: Figure 2
