Abstract:
PURPOSE: To obtain a catecholestrogen glycoside having durable inhibitory action on formation of peroxide lipid, improved water solubility and stability.
CONSTITUTION: A catecholestrogen glycoside of formula I (R1 is H, glucosyl, etc.; R2 is H, glucosyl, etc.; R3 is hydroxyl; R4 is H or ethynyl or R4 and R3 show 0 together; R5 is H or hydroxyl; one of R1 and R2 is glycosyl) such as 1-0-(3-17β-dihydroxy-1,3,5(10)-estratrien-2-yl)-β-D-glucopyranoside. The compound of formula I, for example, is obtained as a desired 2-glucosyl derivative of 2-hydroxy-estradiol by reacting a 2-hydrox yestradiol of formula II protected at the 3- and 17-positions with acetobromoglucose of formula III in the presence of cadmium carbonate and eliminating the protecting group.
