Abstract:

PROBLEM TO BE SOLVED: To measure the peroxidase activity with high sensitivity by employing a reaction mixture obtained through reduction of A): N,N'-di substitution-9,9'-bis dihydroacridine derivative or B): N,N'-di substitution-9,9'-bis acrridium salt of specified structure.
SOLUTION: The chemical emission reagent for measuring the peroxidase activity under presence of hydrogen peroxide is composed of a reaction product obtained through reduction of A having structure shown by formula I (in the formula, R1-R6 represent respective substituents) or B having structure shown by formula II (in the formula, R1-R6 represent respective substituents, Xn represents an n-valent anion, n is 1 or 2). For example, N,N-dimethyl formamide solution of B, i.e., lucigenine, is dispersed into tetrahydrofuran and reduced by hydrogenated lithium aluminum to produce a chemical emission reagent containing N,N'-dimethyl-9,9'-bis dihydroacridine. Activity of horseradish peroxidase can be measured at various concentration levels using this reagent.
