PURPOSE: To obtain a novel compound which is a squalene synthase inhibitor, is useful as a cholesterol-lowering agent or also as an antifungal agent, is also an inhibitor for farnesyl protein transferase and the farnesylation of oncogenic proteins, and is effective in the treatment of cancer.
CONSTITUTION: There is provided a compound of formula I (wherein R is p- hydroxybenzyl, benzyl, or formula II; Z1 to Z3 are each H, a 1-5C alkyl or a 1-5C alkyl substituted with a phenyl not substituted or substituted with methyl, methoxyl, Cl, Br, I, F or OH) or a pharmaceutically acceptable salt thereof. This compound can be obtained by culturing Trichoderma viride MF5628 (ATCC74084) or a variant thereof under conditions favorable for the formation of the compound and isolating the compound from the culture. The compound can be used in combination with a pharmaceutically acceptable nontoxic actionic polymer which can bind with bile acid in a form not re-adsorbed by the gastrointestinal tract (e.g. cholestyramine) and can be administered together with another cholesterol-lowering agent (e.g. HMG-CoA reductase inhibitor).
