Abstract:

PURPOSE: To provide a method for heightening the cleavage of an acyl thiohydantoin bond to use, e.g. for C-terminal peptide sequencing.
CONSTITUTION: An acyl thiohydantoin is alkylated to form an addition product on the thiohydantoin, and the thiohydantoin containing the addition product is brought to react at the acyl bond under a virtually anhydrous acid condition with a cleaving agent and to be cleaved. The product cleaved in C-terminal amino acid sequencing can be analyzed for identifying the C-terminal amino acid.
