CLONING, EXPRESSION AND UTILIZATION OF ACETYL XYLAN ESTERASE FROM FUNGAL ORIGIN

Abstract:

PURPOSE: To obtain the subject recombinant DNA fragment useful for the production, etc., of an acetyl xylan esterase, coding for a protein having the acetyl xylan esterase activity and useful for the xylan hydrolysis, etc., of a feed or a pulp.
CONSTITUTION: A fungus such as Aspergillus niger DS16813 is grown overnight to collect a mycelium, which is then disintegrated with a micromembrane separator. A DNA is extracted with a buffer solution containing phenol, etc., precipitated by an ethanol precipitating method, isolated, treated with a restriction enzyme and joined to a vector to prepare a genomic library, which is further cloned to recover a plasmid containing a DNA coding for a protein having the acetyl xylan esterase activity. The recovered plasmid is then treated with a restriction enzyme and joined to an expression vector to provide a recombinant DNA fragment, which is subsequently inserted into a microorganism host cell to transform the host cell. The transformant is then cultured to recover the resultant product from the resultant cultured product. Thereby, the protein having the acetyl xylan esterase activity is obtained.
