Abstract:
PURPOSE: To determine the quantity of substance to be measured in a specimen by measuring the quantity of labeled substance in an immune complex produced through reaction of a second antibody specifically bound to FC part of a first antibody, while being fixed to water insoluble carrier, a first monoclonal antibody for the substance to be measured, and a labeled substance to be measured or a derivative thereof.
CONSTITUTION: A substance A to be measured includes thyroid hormone, steroid hormone, hapten antigen, and the like and an monoclonal antibody obtained from mouse or rat is employed. A whole antibody including Fc part is employed as a first monoclonal antibody B while an antibody obtained from same animal species as the antibody B is employed as a second monoclonal antibody D. An enzyme, a fluoro-immunoassay, a luminescent-immunoassay, or a radioactive isotope is employed as a substance for labeling the substance to be measured or a derivative C thereof. Glass beads or plastic beads are employed as water insoluble carriers. A, B, C and D are then reacted each other to form an immune complex on the water insoluble carriers. Quantity of labeled substance in the immune complex is then measured thus determining the quantity of the substance A to be measured.
