Abstract:
PURPOSE: To obtain a two-layered composite laminate that is composed of a silicone elastomer and a macroporous polyethylene slab, contains active medicines increased in permeability through a living membrane without skin irritation and is useful for transmembrane administration. CONSTITUTION: This two-layered composite laminate is obtained by mixing medicines, such as tetrahydroaminoacridine, diphenhydramine, hydrocortisone, and/or female sex hormones, such as estrogen and/or progestin, with a liquid silicone, casting the mixture on a heating plate to form partially cured elastomer, and on the other hand, dissolving the medicines into a solvent mixture of two components of ethanol and Miglyol 840 (made by Dynamite Nobel), pouring the solution into pores of a macroporous polyethylene slab molding, laminating the slab with the elastomer to form a two-layered composite laminated, and cutting the laminate to desired size. The medicines are contained in a quantity of 0.1-50wt.%, preferably 0.2-8wt.%.
