Abstract:

PURPOSE: To provide novel heterotrifunctional compounds as photoaffinity labelling of biomolecules which have water-soluble groups before and after labelling of biomolecule and whose solubility is kept so that the labelling molecules protect free into the solution after immobilized on the surface of the biomolecules.
CONSTITUTION: By treating N-(p-azidobenzensulfonyl) N'-(3-carboxypropionyl) cysteamine with N-hydroxysuccinimide, a heterotrifunctional compound of formula I {wherein X1 represents carbonyl or sulfonyl; Y represents hydrogen, Y', X1Y' (wherein Y' represents hydroxyl, amino or alkoxy); R represents a group of formula II [wherein X2 represents disulfide or methylene; R1 represents amino (-NH-R2), R2 represents hydrogen, carboxyalkanoyl, cyclohexandion derivative (-NH-cyclohexendion), carboxyl derivative or the like; (n') and (n") are each of 1 to 6]}, for example N-(p-azidobenzensulfonyl) N'-(3- succinimidyloxycarboxypropionyl)cysteamine, is obtained.
