PURPOSE: To obtain a large amount of CHIKUSETU (Panax) saponin which is useful for treating thrombosis, stress ulcer and other diseases by inoculating Agrobacterium rhizogenes to stem pieces of sterilized callus of TOCHIBA NINJIN (Panax japonicum C.A.Meyer) to induce hairy roots and culturing the hairy roots.
CONSTITUTION: The rhizome of Panax japonicum C.A.Meyer is made germ-free with 70vol.% ethanol and 10wt.% bleaching powder solution, rinsed with sterilized water punched with a cork borer, sliced with a scalpel to 2-3mm thickness tissue sections. These small slices are placed on a culture medium, cultured in dark at 25°C for 6 weeks to induce calli. These calli are cultured in the medium in dark at 25°C for 4 weeks and the culture operation is repeated 4 times to develop the adventitious embryos, then the embryos are grown at 25°C under illumination for 6 weeks to prepare an aseptically regenerated seedling. Then, the stem slice of this seedling is inoculated with Agrobacterium rhizogenes (ATCC 15834), allowed to stand in an agar medium to effect culture at 25°C for 30 days in dark to induce hairy roots and these hairy roots are subjected to shaking culture at 25°C for 60 days to obtain the Panax saponin.
