Abstract:
To provide Shiga toxin A Subunit-derived polypeptides and cell-targeting molecules, comprising amino acid substitutions which impart de-immunization, reduced protease-cleavage sensitivity, and/or a heterologous epitope cargo to the polypeptides, while retaining a Shiga toxin function, such as potent cytotoxicity.SOLUTION: Provided is a cell targeting molecule comprising: i) a binding region capable of specifically binding to an extracellular target biomolecule physically attached to the cell surface of the cell, the extracellular target biomolecule being HER2/neu/ErbB2, PDL1, CD38 or CD20; and ii) a Shiga toxin effector polypeptide comprising a specific amino acid sequence, the amino acid sequence comprising the amino acid substitutions of S45I, V54I, R55L, I57F, P59F, E60T, E61L, G110A, R188A, C242S, R248A and R251A.SELECTED DRAWING: None
