Abstract:

PURPOSE: To enable the production of a polypeptide by the genetic engineering, by using a DNA coded with the amino acid sequence of the polypeptide of human renin, and determining the amino acid sequence.

CONSTITUTION: An intracellular DNA is separated from the fragment of kidney extracted from a patient having high plasma renin activity, and a plasma integrated with the DNA coded with the amino acid sequence of the polypeptide of the DNA is produced. Escherichia coli is transformed with the plasmid to obtain the cDNA library of human kidney. The cDNA is separated from the obtained strain to make a restriction enzyme incision map and determine the base sequence. As a result, the sequences of the 1,218 peptide translation regions (in terms of the number of the base) and 190 nontranslational regions following the 42 nontranslational regions were determined.
