Abstract:

PURPOSE: To make it possible to measure superoxide radical with more higher sensitivity by making the superoxide radical react with pyrido-pyridazine derivative or the salt thereof, and performing chemical light emission.
CONSTITUTION: Superoxide-radical (02-) and pyride-pyridazine derivative or the salt thereof expressed by the formula are made to react, and chemical light emission is generated. In the formula, R1 indicates hydrocarbon or hetero- ring group, which can be substituted, respectively. R2 indicates hydrogen, hydroxyl group, thyol and the like R3 indicates hydrogen atoms, oxygen atoms and hydroxyl group, which can be substituted X indicates oxygen atoms or sulfur atoms. As a sample, which is applied in this method, it is preferable to apply the sample in the measurement of 02-, which is produced from, e.g. cells. As the cells producing 02-, there are the leukocyte-based, vascular-system-based and nervous-system-based cells and the like. When this method is used, the specific light emitting quantity and background light emitting quantity are high, 02- can be detected with high sensitivity and the quantity can be determined.
