PURPOSE: To sensitively and rapidly detect the subject Mycoplasma as the organism causative of human atypical pneumonia using a simple operation by amplifying a DNA region defined by a specified restriction map and specific to the Mycoplasma or a part thereof and carrying out the subject detection using a probe.
CONSTITUTION: A sample containing Mycoplasma hyopneumoniae VPP 11 strain as a pathogenic Mycoplasma is cultured at 37°C and the cell bodies are separated by ultracentrifugation. Genome DNA is separated by the conventional method and treated with a restriction enzyme. A DNA region defined by a restriction map of the formula and specific to the Mycoplasma or a part thereof is amplified by PCR(Polymerase Chain Reaction) method using a specified primer for amplification of the DNA sequence specific to the Mycoplasma and the objective detection of the Mycoplasma is then carried out by using a probe capable of hybridization with the DNA sequence represented by the formula and specific to the Mycoplasma.
