PURPOSE: To determine a bio-substance accurately, by treating a specimen with a specific enzymatic system, eliminating ammonia already exsiting in the specimen, treating it with an enzymatic system which is reacted with the bio-substance to form ammonia.
CONSTITUTION: A specimen is blended with glutamic dehydrogenase, 2-oxoglutaric acid, a small amount of reduction type nicotinamide adenine dinucleotide phosphate (NADPH for short), and an anzyme and substrate to reduce nicotinamide adenine dinucleotide phosphate (NADP+ for short). Ammonia already existing in the specimen is converted into glutamic acid and water, and NADP+ prepared in the conversion is recoverted into NADPH. The specimen is reacted with NADPH and an enzyme which is reacted with the bio-substance to form ammonia, so that the bio-substance is determined accurately.
