Abstract:
PURPOSE: To provide a transdermal delivery device that has a coating layer containing a pressure-sensitive adhesive material, estradiol and a transdermal permeation enhancer comprising a combination of specific compounds on the surface of a flexible substrate and enable a high transdermal permeability and can maintain a desired plasma level of estradiol. CONSTITUTION: On at least a part of the surface of a flexible substrate, is formed a coating layer containing (A) a dermatologically acceptable pressure- sensitive adhesive material, for example, a silicone or polyterpene, (B) estradiol distributed in a substance and (C) a covering layer containing a permeation enhance comprising (C1 ) an unsaturated 12-18C fatty acid and its alkyl ester and 5-30wt.% of glycerol and a 3-6C alkylene-1,2-diol (preferably propylene glycol). In addition, a protecting layer is formed releasably on the opposite side of the coating layer thereby providing the objective transdermal delivery device. In a preferred embodiment, estradiol is contained in the coating layer in a concentration over its solubility.
