Abstract:
Dibenzorhodamines (II), their nucleoside and nucleotide conjugates, and conjugates with other dyes for energy transfer, and 4-aminonaphth-2-ols and their 1-acyl analogues (I), and uses of (II) as fluorescent labels, e.g., for DNA sequencing, oligonucleotide hybridization, polymerase chain reaction detection, and immunoassays. Dibenzorhodamines of formula (II), optionally substituted on the amine groups and/or on the rings by aryl groups, are new: [Image] Independent claims are also included, for (1) conjugates of (II) with nucleosides and nucleotides (NUC-II); (2) energy transfer dye conjugates of (II) comprising: (i) a donor dye capable of absorbing light at a first wavelength and emitting excitation energy in response; (ii) an acceptor dye capable of absorbing the excitation energy emitted by the donor and fluorescing at a second wavelength in response; and (iii) a linker connecting the donor and acceptor dyes, to facilitate efficient transfer of energy between them; in which at least one dye has a formula as (II); (3) A method of polynucleotide sequencing; and (4) Intermediates useful for the synthesis of (II) comprising 4-aminonaphth-2-ols and their 1-acyl analogues of formula (I), optionally substituted on the amine groups, and/or on the rings by aryl groups: [Image] R 1>H or C(O)-Y; and Y : H, 1-8C alkyl, 2-8C alkenyl or alkynyl. phenyl or other monocyclic or polycyclic aromatic, heterocyclic, or a water solubilizing or linking group; provided that, when R 1>H, then a fused 4-7 membered ring is at ring positions C-12 and C-13, and/or C-1 and C-2.
