Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new derivative consisting of a specific digoxigenin derivative containing carbodiimide group, useful as a determination label for nucleic acid detection method or immunoassay to enable highly sensitive analysis by a simple labeling operation and enabling the labeling of a natural nucleic acid.
SOLUTION: This new digoxigenin derivative containing carbodiimide group is expressed by formula I {Z is a digoxigenin-containing group expressed by formula II [A is O, COO or NHCOO; L is an alkylene which may contain a group such as NHCO, CONH, O, S, NR (R is an alkyl), COO or OCO in the main chain; W is quaternary ammonium group; Y1 and Y2 are each a straight- chain bond or same as L; B is H or a univalent organic group same as or different from a group of formula W-Z}. The derivative is useful as a labeling substance in nucleic acid detection method, immunoassay, etc. The compound can be produced by reacting a carbodiimide compound of formula III [W' is a (substituted) amino group] with a halogenated digoxigenin derivative expressed by Z-X (X is a halogen).
