Abstract:

PROBLEM TO BE SOLVED: To provide a new amino terminal propepticle that comprises an intact trimer of procollagen type I isolated without a proteolytic enzyme and is useful as an immunogen for preparation of an antibody for immunoassay of this N- terminal-propeptide.
SOLUTION: This is a mew α1-homotrimer type amino terminal propeptide that is an intact trimer of the procollagen of type I isolated with no use of proteolytic enzyme. This is useful as an immunogen for producing an antibody to be used in the immunoassay of this amino terminal propeptide and permits the determination of the propeptide of the type I procollagen without the inconvenience that is accompanied by simultaneous determination of its monomer form. This propeptide is obtained by saturating the raw material containing this propeptide by ≤50% saturation with ammonium sulfate, subjecting the precipitate to anionic ion exchange chromatography at a pH of 6.5-7.5, eluting the fractions by the concentration gradient of sodium chloride and subjecting the active fractions to the reverse phase separation at a pH of 6.5-7.5.
