PURPOSE: To obtain a new DNA coding for a membrane protein of an avian macrophage having a specific amino acid sequence and creating a bacterium- resistant chicken in relation to the ionic permeability of a macrophage participating in the avian intracellular invasive bacterium resistance.
CONSTITUTION: This new DNA codes for an amino acid sequence of amino acid Nos. 1 to 482 in an amino acid sequence represented by the formula and concerned in the ionic permeability of a macrophage participating in the resistance to an avian intracellular invasive bacterium and further creating a chicken having the resistance to the intracellular invasive bacterium by utilizing the ionic permeability. The DNA is obtained by homogenizing the avian spleen, separating an mRNA according to a conventional method, preparing a cDNA library with the mRNA as a template, screening the prepared library using a probe comprising a part of a membrane protein gene of a murine macrophage, selecting a positive clone, recovering the DNA and carrying out the treatment with a restriction enzyme.
