Abstract:

PURPOSE: To improve the PEPC-productivity of a bacterial cell, by forming a DNA fragment containing phosphoenolpyruvic acid carboxylase-producing gene originated from Corynebacterium genus strain capable of producing glutamic acid.

CONSTITUTION: Whole DNA is extracted from the cell of a Corynebacterium genus strain capable of producing glutamic acid, and the DNA is cut with various restriction enzymes to obtain DNA fragments containing the gene coding phosphoenolpyruvic acid carboxylase (PEPC-producing gene). The restriction enzyme used in the cutting of whole DNA is the one capable of properly cutting the whole DNA of the above bacterial strain, used in a host vector system and usable for the cleavage of vector. The glutamic acid-producing Corynebacterium strain is Gram-positive, immotile, aerobic, non-sporulating and biotin-requiring coryne-type bacterium.
