Abstract:

PROBLEM TO BE SOLVED: To obtain a heat-stable DNA polymerase composition excellent in amplification efficiency of nucleic acid.
SOLUTION: This composition comprises a modified heat-stable DNA polymerase (first DNA polymerase) having 0-5% 3'-5'-exonuclease activity in comparison with a DNA polymerase having 3'5' exonuclease activity before modification and a modified heat-stable DNA polymerase (second DNA polymerase) having 100-6% 3'5' exonuclease activity in comparison with the heat-stable DNA polymerase having 3'-5'-exonuclease activity and the heat-stable DNA polymerase having 3'5'-exonuclease activity before the modification. The first DNA polymerase and the second DNA polymerase have a DNA synthesis rate of at least 30 bases/second and heat stability capable of maintaining ≥60% remaining activity at pH8.8 at 95°C for 6 hours treatment. A long-term nucleic acid can be amplified by using the composition.
